目的研究MEPE/OF45蛋白(Matrix Extracellular PhosphoglycoprotEin/Osteoblast/Osteocyte Factor 45,MEPE/OF45)的功能,制备MEPE/OF45蛋白抗体并检测其定位。方法通过分子克隆方法体外表达MEPE/OF45蛋白片段,获得大量纯蛋白后免疫小鼠并得到抗体,利用抗体进行免疫荧光染色方法检测MEPE/OF45蛋白在细胞内定位情况。结果得到特异性较高的抗体,并观察到MEPE/OF45蛋白在辐射敏感细胞与抗辐射细胞内定位不同。结论MEPE/OF45蛋白不同辐射敏感型的细胞中定位不同,说明MEPE/OF45蛋白参与细胞辐射后DNA损伤修复过程。
Objective To explore the function of matrix extracellular phosphoglycoprotein/osteoblast/osteocyte ( MEPE/OF45 protein) for its antibody preparation and localization. Methods The purified proteins were obtained through MEPE/OF45 protein fragment expressed in vitro by molecular cloning to immunize mice for antibodies to detect MEPE/OF protein localization with immunofluorescence staining. Results The higher specific antibody was obtained, and there was different localization in MEPE/OF45 protein between radiation sensitivity cell and anti-radiation cell. Conclusions MEPE/OF45 protein in different radiation sensitivity cells has different localization, presenting that it participates in cellular DNA injury repairing after radiation.
