[目的]探讨常氧下低氧诱导因子1α(HIF-1α)在传代培养LoVo细胞中的表达。[方法]在常氧下传代培养LoVo细胞,MTT法检测细胞增殖,绘制生长曲线,Western blot检测传代后不同时间点HIF-1α蛋白表达。[结果]细胞生长曲线示LoVo细胞在传代后24h逐渐增加,72h后迅速增加。Westernblot示HIF-1R蛋白表达在传代后24h最高,随时间延长,表达量逐渐降低。[结论]常氧下HIF-1α在LoVo细胞中可低水平表达,传代可引起HIF-1α表达增高,HIF-1α 在促进细胞损伤的修复和细胞增殖中起重要作用。
[Objective]To explore the expression of hypoxia inducible faetor-1α (HIF-1α) under the condition of normoxia and passaged LoVo cells. [ Methods]LoVo cells were cultivated and passaged under normoxia. MTT was used to detect cell proliferation and growth curve was made. Western blot was performed to analyze the protein expression of HIF-1α after passage at different time points. [Results]The number of LoVo cells was increasing at 24 h after passage and in a rapid way at 72 h after passage. The expres- sion of HIF-1α protein was at the highest level at 24 h after passage, and was decreasing with the extension of time. [Conclusions] The expression of HIF-1α was at lower level under normoxia in LoVo cells, which was increased by passage. HIF-1α may p1αy an important role in promoting damaged-cell repairmen and cell proliferation.
