目的:研究束缚-浸水应激不同时间段(O,1,2和4h)雄性Wistar大鼠下丘脑前部(主要是室旁核和视上核)、延髓内脏带(主要是迷走复合体)中突触膨体素和突触蛋白I表达量的变化情况,以期探讨在束缚.浸水应激致胃粘膜损伤过程中上述部位的突触可塑性是否发生了改变。方法:随机将雄性Wistar大鼠分为4组:束缚-浸水应激0,1,2和4h组,利用免疫组织化学和WesternBlot两种方法统计各组下丘脑前部、延髓内脏带中突触膨体素和突触蛋白I的分布和含量变化情况。结果:在束缚-浸水应激0h到4h过程中,下丘脑前部中突触膨体素表达在不同应激时间段差异均无统计学意义,但突触蛋白I,尤其是突触蛋白Ib的含量发生了显著性变化(P〈0.05);迷走复合体中的突触膨体素和突触蛋白I的表达均发生了显著性变化(P〈0.05)。结论:这些结果提示,下丘脑前部、延髓内脏带在束缚.浸水应激致胃粘膜损伤的中枢调控过程中均发生了突触可塑性的变化。
Objective: In order to explore whether the synaptic plasticity in the anterior hypothalamus [ paraventricular nucleus (PVN) and supraoptic nucleus (SON) ] and the medullary visceral zone [ the dorsal vagal complex (DVC) ] was changed during the different durations of restraint water-immersion stress (RWIS) in rats, the expression and amounts of synaptophysin (SYN) and Synapsin I were determined. Methods: Male Wistar rats were randomly divided into 4 groups designated according to their duration of RWIS, respectively 0 h, 1 h, 2 h and 4 h. Additionally, immunohistochemistry staining technique and Western Blot technique were used to determine the expression and amounts of SYN and Synapsin I in the anterior hypothalamus and the medullary visceral zone during RWIS. Results: During RWIS 0 h to 4 h, amounts and distributions of Synapsin I, especially Synapsin I b, was significantly different in the anterior hypothalamus with dif- ferent durations of RWIS ( P 〈 0.05 ), but SYN didn't changed. In the medullary visceral zone, the amounts and distribu- tion of SYN and Synapsin I were all significantly changed ( P 〈 0.05 ). Conclusion: These data indicate that the synaptic plasticity is changed in the anterior hypothalamus and the medullary visceral zone in rats exposed to RWIS.
