以超级稻品种两优培九成熟胚为外植体,研究了外源激素和化学试剂对超级稻离体再生培养的影响.结果表明,成熟胚在MS+2,4-D2mg/L+KTlmg/L培养基上,愈伤组织诱导率达94%;愈伤组织在MS+6-BA2mg/L+KTlmg/L+NAA0.5mg/L培养基上,分化率为76.66%;当分化培养基中添加5mmol/LCuS04,愈伤组织生长旺盛,分化率提高,可达86.66%.
Using mature embryos of super rice variety, Liangyoupei 9, as explants, the effects of exogenous hommones and chemic reagent on the cellar culture of the super rice in vitro were investigated. The results showed that the induction frequency of mature embryos callus on MS+2,4-D 2 mg/L + KT lmg/L medium was 94%; the differentiation frequency of buds on MS+6-BA 2 mg/L+KT 1 mg/L+NAA 0.5 mg/L medium, used as differentiation medium, was 76.66%; When 5 mmol/L CuSO4 was added to the differentiational medium, it reached as high as 86.66% and the callus grew well.
