为进一步研究超级稻的生长素结合蛋白,采用生物信息学方法获得其全长cDNA序列,并进行分析;对全长序列进行了全长验证。从数据库搜索获得了一条水稻cDNA序列,含一个推测的206个氨基酸的开放阅读框,与其它植物的ABP1cDNA序列和ABP1蛋白质序列具有明显的同源性,并且其推测编码的蛋白质的各种性质。类似于其它植物的ABP1。结果显示,该序列为水稻的ABP1cDNA。以该序列为指导,利用RT—PCR扩增超级稻ABP1cDNA,并克隆测序,其序列与电子搜索结果一致。
In order to study super rice auxin-binding protein 1, its full-length cDNA sequence was obtained and analyzed by bioinforrnatics and laboratory bench-work. In the results, a rice cDNA sequence was got from nucleotide sequence database, the putative open reading frame of the sequence encodes a presumptive protein containing 206 amino acid residues. Blast against the current GenBank DNA and protein sequence database reveal significant homology with the ABP 1 of other plants, and the functions of presumptive protein resemble these of the ABP 1 of other plants. These results suggest the sequence is rice ABP1cDNA. Based the sequence, ABP1cDNA of super rice was amplified with RT-PCR and cloned for sequencing, the result of sequencing accord with electronic seaching outcome.
