目的 评价Homer家族在注意缺陷多动障碍(ADHD)动物模型幼年自发性高血压大鼠(SHR)及其对照Wistar京都大鼠(WKY)海马中的表达差异,分析其与ADHD患儿学习记忆功能障碍的关系。方法SHR大鼠12只、WKY大鼠11只,平衡数天,断头处死,取其海马组织。取8只SHR大鼠及7只WKY大鼠海马组织,利用反转录聚合酶链反应(RT—PCR)技术检测SHR/WKY大鼠海马组织中Homerla、Homerlb/c、Homer2a/bmRNA的表达。取其余4只SHR和4只WKY大鼠海马组织,利用Westernblot技术检测在2组大鼠海马组织中存在差异表达的上述基因蛋白在SHR/WKY大鼠海马中的表达。结果1.SHR大鼠Homer 1a、Homer2a/bmRNA表达量均较WKY大鼠显著降低(t=2.95P〈0.01;t=2.66P〈0.05),Homer 1b/c mRNA表达水平在2组间无显著差异(t=0.74P〉0.05)。2.SHR大鼠海马组织中Homer 1a、Homer2a/b蛋白表达量均较WKY大鼠显著降低(t=4.31,3.27P〈0.01).结论Homer 1a Homer,2a/b可能在ADHD发病机制中发挥重要作用。

Objective To investigate the differential expressions of several Homer isoforms in hippocampus of spontaneous hypertensive rat and Wistar- Kyoto rats (SHR/WKY) ,and the most frequently used animal model of attention deficit hyperactivity disorder (ADHD), and discuss the relationship between Homer and ADHD. Methods Twelve cases of SHR and 11 cases of WKY rats were allowed some days to acclimatize, then sacrificed to dissect the hippocampus. Total RNA of hippocampus of SHR ( n = 8 ) and WKY ( n = 7 ) rats were isolated to detect the differential expressions of Homer 1a, Homer 1 b/c, Homer 2a/b mRNA using reverse transcription polymerase chain reaction. Protein concentrations of hippocampus of the left 4 rats in each group were isolated to detect the expression of these genes protein which were found expressed differentially on mRNA level, using Western blot. Results 1. The expressions of Homer 1 a and Homer 2a/b gene were significantly lower in hippocampus of SHR than those in WKY group ( t = 2. 95 P 〈 0.01 ; t = 2. 66 P 〈 0.05 ). There was no difference in the expres- sion of Homer 1b/c gene in hippocampus between SHR and WKY groups (t = 0. 74 P 〉 0.05 ). 2. The expressions of Homer 1 a and Homer 2a/b protein were significantly lower in hippocampus of SHR than those in WKY ( t = 4.31,3.27 Pa 〈 0.01 ). Conclusion Homer 1a and Homer 2a/b may play an important role in pathogenesis of ADHD.
