目的 探讨体外研究毛囊被剪成不同大小的移植物后对其活性的影响.方法 将显微镜下分离所得到的含有单个毛囊的移植物,按照其外根鞘周围留有保护组织厚度的不同分为A(0mm)、B(0.3mm)、C(0.6mm)3组,培养6 d后观察其活性情况,并用毛干生长率、毛干生长长度和毛囊的组织形态学变化来衡量.结果 A、B、C 3组中毛干生长率分别为81.5%、91.7%,92.6%,A和B组与A和C组间的差异有统计学意义(P<0.0125,P<0.0125),B和C组间的差异无统计学意义(P>0.0125);毛干的平均生长长度分别为(1.34±0.10)、(1.36±0.95)、(1.36±0.12)mm.组间均数的差异均无统计学意义(P>0.05);各组间毛囊的细胞形态无明显变化.结论 毛囊外根鞘周嗣留有0.3 mm厚度的保护组织,既不影响其活性,又可以增加单次移植的密度.本实验结果 为临床毛发移植提供了参考数据.
Objective To observe viability of trimmed different size of hair follicles by quantitative methods in vitro. Methods Firstly, human hair follicles were isolated by microdissection in 9 patients. Secondly, the hair grafts were divided into group A (0 mm), B (0.3 mm) and C (0.6 mm) based on thickness of retained protective tissue around the outer root sheath. Thirdly, all hair follicles were carried out the organ culture. Fourthly, at the sixth day, some were randomly picked up to judge their viability by calculating growth rate, measuring hair elongation and observing morphologic changes. Results The survival rate of the three group was 81.5%, 91.7%, 92.6% respectively and the mean 6-day hair elongation was ( 1.34 ± 0. 10), ( 1.36 ± 0.95 ), ( 1.36 ± 0.12) mm, respectively. There were all significant differences among A and B ( P 〈 0. 0125) or A and C ( P 〈 0. 0125), but reversed between B and C ( P 〉 0. 0125 ). No statistically signifi- cant differences were found among the hair elongation of follicles from A, B and C groups ( P 〉 0. 05). No alteration were detected in follicles among there groups. Conclusion The viability is not influenced when hair follicle retained 0. 3 mm thickness protective tissue around the outer root sheath in vitro experiment. These resuits lay reference data for hair transplantation in clinic.
