目的:探讨转化生长因子-β的两种异构体(TGF—β1和β2)及其受体(TGF—ORII)在黑毛白皮小香猪皮肤及毛囊中的表达特征。方法:用免疫组化方法确定TGF-D亚型及其受体在黑毛白皮小香猪皮肤及毛囊中的定位和表达量的变化规律。结果:TGF-β蛋白颗粒主要分布于表皮角质形成细胞、生长后期及退行期的毛囊外根鞘和内根鞘细胞,在皮下脂肪细胞中亦有弱阳性表达。TGF—β2的蛋白颗粒在表皮角质形成细胞有阳性表达,在真皮乳头层大部分成纤维细胞和血管内皮细胞中均有强阳性表达。在毛囊的退行期,毛囊外根鞘细胞、毛隆突细胞和结缔组织鞘成纤维细胞中有阳性表达。TGF-β R Ⅱ主要存在于表皮角质形成细胞,在毛囊结缔组织鞘中部分成纤维细胞和外根鞘细胞中有阳性表达,特别是在退行期早期的毛乳头基质细胞、外根鞘细胞中有强阳性表达,在退行中晚期的外根鞘部分细胞中有阳性表达,但明显减弱。TGF-β1、TGF—β2和TGF—β R Ⅱ在猪皮肤中蛋白表达量分别为17.0±2.5,24.2±3.1,13.2±2.9。结论:TGF—D的异构体及其受体主要表达于毛囊生长期晚期和退行期早期,可能在毛囊从生长期向退行期转化的过程中起重要的作用。

Objective To observe the localization and expression characteristics of two transforming growth factor-β (TGF-β) isoforms,and their receptor in skin and hair follicles of Tibet mini-pigs with dark hair in order to explore their potential biological significance. Methods Protein expression of two TGF-15 isoforms and their receptors TGF-βR Ⅱ were respectively observed with immunohistochemical method in 5 skin specimens. Results Positive immunohistochemical signals of TGF-β could be found in cytoplasm of epidermal keratinocytes, outer and internal root sheath cells of late anagen and catagen hair follicles tissue and subcutaneous fat cells. Protein expression of TGF-β2 was higher in epidermal keratinocytes, dermal most fibroblasts and vascular endothelial cells. During catagen of hair follicles, TGF-β2 protein could be found in outer root sheath cells, follicle bulge cells and fibroblasts of connective tissue sheath. TGF-β RⅡ was mainly distributed in the epidermal keratinocytes, outer root sheath cells and part fibroblasts of connective tissue sheath. During early catagen, TGF-βR Ⅱ was especially strongly expressed in hair papilla matrix cells and outer root sheath cells, and could still be found in midanaphase of catagen, but was obviously weakened. The expression levels of TGF-β1, TGF-β2 and TGF-βR Ⅱ proteins were 17.0±2.5, 24.2±3.1 and 13.2±2.9 respectively. Conclusion The two transforming growth factor-β (TGF-β) isoforms and their receptor TGF-13R II were mainly expressed during late anagen and early catagen. It is suggested that endogenous TGF-β1 and TGF-β2 and their receptor might play important role in the process of hair cycle transition from anagen to catagen.
