目的:研究未折叠蛋白反应对顺铂诱导肝癌细胞凋亡的影响及其机制。方法:在采用二硫苏糖醇和衣霉素诱导未折叠蛋白反应的基础上,利用流式细胞和免疫印迹技术分析未折叠蛋白反应对顺铂诱导的肝癌细胞凋亡的影响及其机制。结果:二硫苏糖醇和衣霉素预处理促进顺铂诱导的自噬反应并抑制顺铂介导的肝癌细胞凋亡,抑制自噬反应明显削弱二硫苏糖醇和衣霉素预处理对顺铂作用下肝癌细胞的保护作用。结论:未折叠蛋白反应能够通过促进自噬抵抗顺铂诱导的肝癌细胞凋亡。
Objective: To investigate the role and molecular mechanism of unfolded protein response on cisplatin-induced apoptosis in human hepatocellular carcinoma cell.Methods: Dithiothreitol and tunicamycin were used to establish unfolded protein response microenvironment in SMMC-7721 and HepG2 cells.The effects of unfolded protein response on cisplatin-induced SMMC-7721 and HepG2 cells’ apoptosis were analyzed by flow cytometry and Western blot analysis.Results: Dithiothreitol and tunicamycin preincubation obviously increased cisplatin-mediated autophagy and inhibited cisplatin-induced apoptosis in SMMC-7721 and HepG2 cells.Autophagy suppression inhibited the protective effect of unfolded protein response precondition in cisplatin-treated SMMC-7721 and HepG2 cells.Conclusion: Optimum unfolded protein response protects HCC cells against cisplatin-induced apoptosis through autophagy regulation.
