以烟草悬浮细胞BY-2为材料,探讨细胞外ATP对ZnCl2诱导的细胞活性降低和呼吸抑制的调节作用.结果表明,随着ZnCl2浓度逐渐增加,细胞活性、胞外ATP浓度和细胞呼吸速率均随之降低.在400mmol/LZnCl2胁迫细胞的条件下,研究了细胞外ATP对ZnCl2诱导的细胞活性降低和呼吸抑制的调节作用.结果发现,与400mmol/LZnCl2胁迫下的细胞相比,加入20μmol/L外源ATP到Zn-Cl2胁迫下的细胞中,会使细胞的活性、胞外ATP浓度和细胞呼吸速率均有显著升高.胞外ATP浓度的变化可能是造成ZnCl2胁迫下细胞活性受抑制的原因之一,同时胞外ATP对ZnCl2胁迫下细胞的活性具有一定的调节作用.
Tobacco BY-2 suspension cells were used to study the effect of the extracellular ATP on the decreasing the cell activity and respiration rate induced by ZnCl2 stress. The results showed that, with the increment of the concentrations of ZnCl2, the cell activity was decreased, the same as extracellular ATP and respiratory O2 uptake. The role of extracellular ATP in the decrease of ZnCl2-induced cell activi- ty and respiration rate was studied in the cell subjected to 400 mmol/L ZnCl2. And the results showed that, with the addition of 20 μmol/L exogenous ATP, ZnCl2 stress induced the decrease of cell activity, and extracellular ATP and respiratory O2 uptake were both increased. These results suggested that the change of extracellular ATP might be one of the reasons behind ZnCh stress- induced inhibition of cell activity, and extracellular ATP could function in mediating the ZnCl2-induced cell death.
