目的应用蛋白质组学技术研究强化铁营养对缺铁大鼠耳蜗膜性组织肌动蛋白(actin)、肌球蛋白(myosin)、结蛋白(desmin)和热休克蛋白(heat shock protein,HSP)表达的影响。方法体重25~35g、健康、无耳疾、ABR阈值正常的1~2周龄Wistar大鼠120只,随机分成正常对照组20只,喂以标准饲料16周;缺铁大鼠组100只,喂以缺铁饲料8周后,复制出至少有一耳ABR阈值改变≥15dB的大鼠共41只,再将该41只大鼠随机分成缺铁耳聋组20只和强化铁治疗组21只,缺铁耳聋组继续喂以缺铁饲料8周、强化铁治疗组喂以强化铁饲料8周后,取大鼠耳蜗膜性组织进行蛋白质组学分析,对各组间actin、myosin、desmin和HSP表达的差异进行比较。结果缺铁耳聋组大鼠耳蜗膜性组织中的actin、myosin及HSP种类较正常对照组减少,并出现特异蛋白质desmin;强化铁治疗组大鼠耳蜗膜性组织myosin和HSP种类较缺铁耳聋组增加,actin种类未见明显改变,desmin消失。结论铁缺乏导致的耳蜗actin、myosin及HSP种类减少,以及出现特异性蛋白质desmin,可能与缺铁性聋的发生发展有关;强化铁营养可使缺铁耳聋大鼠耳蜗myosin和HSP种类增加,desmin消失。
Objective To study the effects of fortified iron nutrition on actin,myosin,desmin.and heat shock protein expression in membranous cochlea in iron-deficient rats using proteomic strategy.Methods 120 healthy Wistar rats(2535 g body weight,12 weeks old)with normal ABR threshold and no ear disease were randomly divided into two groups.In group A (normal control),20 rats were given the final Fe-supplemented diet for 16 weeks.In group X(experimental iron deficiency),100 rats were fed with the basic Fe-deficient diet for 8 weeks.In group X,41 rats with sensorineural hearing loss induced by iron dificiency were then randomly devided into 2 groups:group B (iron-deficient hearing loss group,20 rats ).and group C (fortified iron nutrition group,21 rats ).The rats in group B were fed with iron deficiency diet for another 8 weeks while group C rats were given the fortified iron nutrition diet for another 8 weeks.The proteins of cochlea were examined for finding the differences and searching for proteomic profile using BIOWORKS software.Results The subtypes of actin,myosin and heat shock protein were decreased in the cochlear of the rats in group B,compared with those of in group A.In group C,the subtypes of myosin and heat shock protein were increased in the cochlear ,compared with those of in the group B.Protein expression changed significantly in the cochlea in iron-deficient hearing loss rats after the treatment of fortified iron nutririon.Conclusion The decrease of actin,myosin and heat shock protein expression,and the increase of desim expression may be responsible for iron deficiency hearing loss.The fortification of iron nutrition could increase the expressions of myosin and heat shock protein in membranous cochlea in the rats with iron deficiency hearing loss.
