以纯化的重组兔出血症病毒核衣壳蛋白(VP60)为抗原,免疫BALB/c小鼠,采用杂交瘤细胞技术制备抗衣壳蛋白的单克隆抗体,获得了2株能稳定分泌抗VP60蛋白单克隆抗体的杂交瘤细胞株(AE11和AH4).分别利用Western blot、间接ELISA和间接免疫荧光(IFA)等方法对单抗的免疫活性和特异性进行检测,结果证明,此两株单抗均能识别重组的VP60和天然病毒的衣壳蛋白,而IFA结果显示只有AE11能和RHDV反应,可观察到强烈的特异性绿色荧光,表明成功制备了抗RHDV的VP60特异性单抗.
BALB/c mice were immunized with the recombinated cap sid protein (VP60) of Rabit hemorrhagic disease virus (RHDV), and anti-VP60 mAb was prepared by using hybridoma techonology. The specificity of mAbs (AEll and AH4) was characterized by Western-blot and indirect enzyme linked immunosorbent assay (ELISA). The results indicated that the mAbs was able to recognize the recombinated protein (rVP60) and the natural viral protein. However,the result of IFA indicated that only AE1l could react to RHDV,and the infected cells were stained clearly. The mAbs against VP60 were prepared successfully,which provides valuable reagent for developing the diagnosis reagent and studies on RHDV.
