目的观察天麻钩藤饮对帕金森病(PD)大鼠神经行为学及氧化应激的影响。方法采用6羟基多巴胺(6-OHDA)注射于脑右侧黑质造成单侧PD损毁模型,并用中药天麻钩藤饮(浓度为5.13g/ml)进行治疗,共给药14d。同时设立正常对照组、假手术组,观察各组大鼠神经行为学的变化,同时运用化学比色法测定大鼠中脑黑质、纹状体部位活性氧、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)的活性。结果神经行为学方面,模型组大鼠治疗前后旋转圈数无显著差异;天麻钩藤饮组大鼠旋转圈数较模型组显著减少(P〈0.05),且治疗前后比较差异显著(P〈0.05);模型组活性氧、MDA明显升高,GSH、GSH-Px、SOD明显降低,与正常对照组、假手术组比较,差异显著(P〈0.05或P〈0.01);而天麻钩藤饮对其均有明显的改善作用(P〈0.05)。结论天麻钩藤饮可以明显改善PD大鼠的神经行为学变化,并可以提高机体的抗氧化和清除自由基的能力。
Objective To observe effect of tianma gouteng drink on the behavioural and oxidation stress response of Parkinson' s disease (PD) rat. Methods Rats were divided into normal control, sham operation, model, and treatment groups (6 rats in each group). PD rats were produced by injection of 6-OHDA stereotaxically into the right side of the mesencephic ventral tegmental area (VTA) and sub- stantia nigra pars eompacta (SNc) and treated with tianma gouteng drink (5. 13 g/ml) for 14 days. Behavioural changes and the activity of reactive oxygen species ( ROS), glutathione ( GSH), glutathione peroxidase ( GSH-Px), superoxidase dismutase ( SOD), malonaldehyde (MDA) of the substantia nigra and corpus striatum of the PD rats were observed. Results There was no significant difference in rotation be- haviour between before and after treatment in model group ( P 〉 0. 05 ). Compared to model group, the rotation behaviour was significantly decreased in treatment group, and there was significant difference between before and after treatment in treatment group (P 〈 0. 05). Com- pared with the normal control and sham operation groups, ROS and MDA contents were obviously increased and GSH, GSH-Px and SOD con- tents were obviously reduced in model group (P 〈 0.05 ). However, the above indicators were improved in treatment group (P ~ 0. 05 ). Conclusions Tianma gouteng drink can significantly improve the neurological behavior, and enhance anti-oxidation ability and eliminate free radicals of PD rats.
