目的 观察音猬因子(Shh)信号通路在小鼠急性胰腺炎中的作用.方法 雨蛙素腹腔注射诱导急性胰腺炎模型后,引入Hedgehog信号的拮抗剂环杷明(Cyclopamine),用逆转录-聚合酶链反应(RT-PCR)、Western blot方法检测Shh在胰腺组织的表达,同时检测了胰腺髓过氧化物酶(MPO)水平.结果 与对照组比较,雨蛙素注射1h后,Shh表达逐渐升高,在24h到达最高.与对照组比较[(231.13 ±10.03) U/mg],MPO活性逐渐升高,6h到达高峰[(1 255.56±126.68) U/mg],24h恢复正常[(230.48±17.69) U/mg].当Cyelopamine抑制Hedgehog信号后,胰腺炎症明显加重.结论 在急性胰腺炎性过程中,Shh起重要的保护作用.
Objective To study the potential role of Sonic Hedgehog (Shh) signal pathway in mice acute pancreatitis.Methods Acute pancreatitis was induced by intraperitoneal injections of cerulean 7 times at hourly intervals in mice,and the specific antagonist of Hedgehog was injected as well.Shh expression was detected by real-time reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry (IHC).The level of pancreatic myeloperoxidase (MPO) was determined.Results As compared with control group,RT-PCR and Western blotting results showed Shh in experimental group was increased,at 24 h,up to peak values.Inflammation was assessed by the activity of MPO.In control group,the MPO activity was (231.13 ± 10.03) U/mg,and experimental group showed a rapid increase in MPO activity,at 6 h,up to peak values [(1 255.56 ± 126.68) U/mg].The MPO activity recovered baseline at 24 h [(230.48 ± 17.69) U/mg].When hedgehog signal was inhibited by cyclopamine,the inflammation of pancreas was more serious.Conclusion During the acute pancreatitis,Shh signal may play a protective role to regulate the pancreatic inflammation.
