目的 观察肿瘤坏死因子受体相关6(TRAF6)在小鼠急性胰腺炎相关的肺损伤中的作用.方法 雨蛙素7次腹腔注射诱导急性胰腺炎模型,用逆转录-聚合酶链反应(RT-PCR)、Western blot方法检测TRAF6肺组织表达.也检测了肺髓过氧化物酶(MPO)的水平.结果 与对照组比较,雨蛙素注射后1h,TRAF6表达明显下调,然后在2、4、12 h开始逐渐恢复,在24 h,表达明显上调(P〈0.05).在胰腺炎症早期,MPO水平明显升高(1h 477.83±30.24 2 h 797.50±34.25 4 h1255.50±126.66),12 h明显下降(608.17±30.27),24 h接近基线水平(230.50±17.69).结论 在急性胰腺炎相关的肺损伤过程中,TRAF6可能参与调节肺炎症进程.
Objective To study the potential role of TRAF6 in mice acute pancreatitis associated lung injury. Methods Acute pancreatitis was induced by intraperitoneal injections of cerulean 7 times at hourly intervals in mice. TRAF6 expression was quantitatively detected by real-time reverse transcription-polymerase chain reaction ( RT-PCR) and Western blotting. The level of myeloperoxidase ( MPO) was determined. Results As compared with saline-induced control mice, RT-PCR and Western blotting results showed TRAF6 in cerulien-induced acute pancreatitis mice was down-regulated in the lung within 12 h, adversely , significant up-regulation could be observed at 24 h (P〈0.05 ). During the early time of acute pancreatitis, the level of MPO was obviously increased (477. 83±30.24 at 1h 797. 50±34.25 at 2 h 1255. 50±126. 66 at 4 h), decreased at 12 h (608. 17 ±30. 27) , restored to basic level at 24 h (230. 50 ±17.69). Conclusion TRAF6 may play an important role in acute pancreatitis associated lung injury.
