目的:观察去势山羊接受静脉注射同源异体骨髓间充质干细胞(Bone marrow mesenchymal stem cells,BMMSCs)后,其体内BMMSCs增殖及分化功能的改变。方法:取3—6岁关中雌性山羊15只,随机分为双侧卵巢切除组(OVX)、双侧卵巢切除24h后静脉注射同源异体BMMSCs组(OVX+injection)和假手术对照(sham)组(n=5),并进行相应处理。1年后分离各组山羊的BMMSCs,MTT法检测其增殖能力;成骨、成脂诱导培养后,采用茜素红染色和油红O染色观察其多向分化能力。结果:去势1年后,OVX组与Sham组相比,BMMSCs的增殖能力和成骨分化能力均明显下降,而成脂分化能力明显增强(P〈0.05);OVX+injection组BMMSCs的增殖和成骨分化能力均较OVX组明显提高,而成脂分化能力明显下降(P〈0.05)。结论:静脉注射同种异体BMMSCS可逆转去势后BMMSCs的功能异常。
AIM : To investigate the effects of intravenous injection of allogeneic bone marrow-derived mes- enchymal stem cells (BMMSCs) on the proliferation and differentiation of BMMSCs in oophorectomized femal goats. METHDOS: 15 female goats aged 3-6 years old were randomly divided into bilateral ovariectomy group (OVX), sham group (Sham) and intravenous injection of allogeneic BYlMSCs 24 h after bilateral ovariectomy( OVX + injection) group. Goats in all groups were sacrificed 1 year after treatment. The proliferation and differentiation of BMMSCs were examined by MTT assay, alizarin red and oil red staining respectively. RESULTS : The capabilities of proliferation and osteogenic differentiation of BMMSc in OVX group were lower than those in the sham group(P 〈 0.05 ) , but the capa- bility of adipogenic differentiation was higher( P 〈 0.05 ). The abilities of proliferation and osteogenic differentiation of BMMSc in OVX + injection group were higher than those in OVX group(P 〈 0.05), and the ability of adipogenic dif- ferentiation was lower(P 〈 0.05). CONCLUSION: The dysfunction of BMMSCs in the oophorectomized femal goats can be reversed by intravenous injection of allogeneic BMMSCs.
