雌激素效应增强在良性前列腺增生(BPH)发生发展中发挥了重要作用.收集20例BPH和10例正常前列腺(NP)组织标本,采用免疫组化染色和免疫荧光方法研究良性前列腺增生组织中上皮细胞角蛋白(CK)、非肌型肌球蛋白(NMMHC)、增殖细胞核抗原(PCNA)和雌激素受体α(ERα)的表达及其定位关系.结果表明,与NP相比,在BPH组织基底细胞中与干/祖细胞相关的CK14和CK5以及与去分化相关的NMMHC和ERα的表达明显增加;而且CK14、NMMHC和ERα有共定位的关系.BPH组织中部分基底细胞去分化为干性细胞;雌激素效应增加使基底细胞发生去分化是BPH发生和发展的重要机制.
Enhanced estrogen effect is believed to play an important role in the pathological benign prostate hyperphasia(BPH).The the expression of epithelium cytokeratin(CK),non-muscle myosin heavy chain(NMMHC),proliferating cell nuclear antigen(PCNA) and estrogen receptor α(ERα) in 20 cases of BPH and 10 cases of normal prostate(NP) tissue samples was studied.Data from immunohistochemical staining showed that the expression of stem/progenitor cells related marker include CK14 and CK5,the dedifferentiation related marker NMMHC,as well as ERα are all increased significantly in the basal cells in BPH tissues.Data also suggest that CK14,NMMHC and ERα have a colocalization relationship,and the colocalization of these three proteins are further proved by immunofluorescence double staining experiment.These results indicate that basal cells dedifferentiated to stem-like cells in BPH tissues and the enhanced estrogen effect induced basal cell dedifferentiation is an essential mechanism for the initiation and development of BPH.
