雌激素在前列腺癌转移中的作用已有报道,但其作用机制尚不清楚.研究了雌激素受体辅助抑制因子REA在前列腺上皮细胞和前列腺组织中的表达及其对前列腺癌细胞生物学功能的影响.发现在前列腺癌细胞和组织中随恶性程度增加REA的表达逐渐增强.划痕愈合实验证实了REA能够明显促进前列腺癌细胞的迁移,而且其促进作用与雌激素和ER存在着协同作用.这些研究结果为揭示雌激素效应及其受体亚型的表达水平在前列腺癌转移中作用机制的探讨奠定了实验基础.
It has been noticed that estrogen could enhance the metastasis of prostate cancer for decades, but the mechanism is remain unclear yet. The role of steroid receptor cofactor REA in regulating the biological behaviors of prostate cancer cells was investigated. Compared to non-cancer prostate epithelial cell lines and tissue samples, the expression of REA was sig- nificantly higher in cancer cell lines and tissue specimens, and the REA level correlated posi- tively with gleason grade in tumor. Over expression of REA in prostate cancer cells could pro- mote cell proliferation and migration, and there is a functional synergy among REA, estrogen and estrogen receptor beta in prostate cancer cells metastasis. It was indicated that REA plays an important role in estradiol's pro-metastasis effect through specific estrogen receptor subtype during prostate cancer progression.
