目的将从双胸蚓组织中纯化的核酸酶EWD2免疫新西兰大白兔制备兔抗EWD2多克隆抗体。方法将纯化的双胸蚓核酸酶EWD2免疫新西兰兔,制备兔抗EWD2血清,以Western Blotting和ELISA方法分析其特异性和效价。结果EWD2蛋白免疫新西兰兔成功获得多克隆抗体。Western Blotting结果显示制备的抗体具有较高的特异性,ELISA显示效价为1:6000。结论获得了双胸蚓核酸酶EWD2多抗血清,为今后对双胸蚓核酸酶活性和结构的研究建立了基础。
Objective To prepare polyclonal anti-EWD2 antibody in New Zealand rabbits immunized with nuclease EWD2 purified from Lumbricus Bimastus. Methods The polyclonal anti-EWD2 antibody was prepared by immunizing New Zealand rabbits with purified EWD2 protein. The specificity and the titer of the antibody were determined by Western blotting and ELISA, respectively. Results The polyclonal anti-EWD2 antibody was successfully obtained. The specificity of polyclonal anti-EWD2 antibody was vali- dated by Western blotting. The titer of polyclonal anti-EWD2 antibody was 1 : 6 000. Conclusion Polyclonal anti-EWD2 antibody can be successfully prepared , which could provide a basis for further study on activity and structure of nucleases in Lumbricus Bimastus .
