目的研究银屑病患者皮损闸充质干细胞(mesenchymal stem cells,MSCs)细胞因子分泌水平。方法分离患者皮损与健康人皮肤MSCs,流式细胞术及多向分化法进行细胞鉴定;ELISA法检测第三代皮肤MSCs培养上清液白细胞介素-8和粒细胞-巨噬细胞集落刺激因子的分泌水平。结果两组MSCs的细胞形态、表面标志和多向分化能力相似,细胞标记CD29,CD90及CD105均呈高表达,而CD34,CD45及HLA—DR呈阴性,均可向脂肪和成骨细胞分化。与健康对照相比,银屑病患者皮损MSCs分泌白细胞介素-8和粒细胞-巨噬细胞集落刺激因子水平均显著增高,差异有统计学意义(P〈0.001)。结论银屑病患者皮损MSCs细胞因子分泌异常,可能是导致皮损微环境改变的重要原因。皮损MSCs异常可能参与银屑病的发病,
Objective To detect the levels of cytokine secreted by skin mesenchymal stem ceils (sMSC) in psoriatic patients. Methods sMSC were separated from psoriasis and normal skin,which were also identified by flow eytometry and muhidirectional differentiation. The levels of GM-CSF and IL-8 were detected by ELISA secreted by the third passage of sMSC. Results The cell morphology, surface markers and differentiation ability were similar in psoriasis and normal. Flow cytometry showed higher expression of CD29, CD90 and CDI05 ,while CD34, CD45 and HLA-DR negatively expressed in sMSC of two teams. In addition, sMSC could be induced into adipogenic cells and osteobJasts in both two teams. The levels of GM-CSF and IL-8 in psoriasis significantly increased compared with normal (P 〈 0. 001 ). Conclusion The abnormality of cytokine secretion by sMSC may result in the change of skin microenvironment in patients with psoriasis,and participate in the pathogenesis of psoriasis,
