开放阅读框3(Open reading frame 3,ORF3)是猪流行性腹泻病毒(PEDV)基因组中的惟一辅助基因。前期研究表明,PEDV疫苗株的ORF3在245~293位缺失49个核苷酸。在ORF3缺失区域两端设计合成引物建立反转录-聚合酶链反应(RT-PCR)可以对PEDV野毒株和疫苗株进行鉴别诊断,野毒株扩增产物为319 bp,疫苗株扩增产物为270 bp。用该鉴别诊断方法对猪传染性胃肠炎病毒、猪轮状病毒、猪瘟病毒、猪伪狂犬病病毒和猪繁殖和呼吸综合征病毒的cDNA进行扩增均无特异性条带,说明该方法特异性强。现地病料的检测结果表明,该鉴别诊断方法能够区分PEDV野毒和疫苗株,而且可以排除其他病毒的影响,有助于减少免疫猪被淘汰的可能,降低经济损失。
Open reading frame 3(ORF3) is the only accessory gene in the porcine epidemic diarrhea virus(PEDV) genome.Previous study indicated that ORF3 of PEDV vaccine strain had a 49-nt deletion at nt245-293.A reverse transcription-polymerase chain reaction(RT-PCR) method was developed to distinguish wild PEDV from the vaccine strain by using a pair of primers.The PCR products of wild strains are of 319 bp in size,while those of the vaccine strain are of 270 bp in size.No PCR products were amplified from the cDNAs of swine transmissible gastroenteritis virus(TGEV),porcine rotavirus(PoRV),classical swine fever virus(CS.FV),porcine pseudorabies virus(PPV) and porcine reproductive and respiratory syndrome virus(PRRSV),respectively.The results indicat that this method has good specificity for PEDV.The results of the detection of field samples indicated that this method can effectively distinguish wild PEDV from the vaccine strain and reduce the economic losses.
