以草莓(Fragaria×ananassa)品种‘花姬’为试材,通过PCR和RACE的方法克隆出LFY同源基因的cDNA全长序列,命名为FaLFY(GenBank收录号JN788260),通过定量PCR的方法检测了该基因在草莓植株各组织和花器官中的表达。结果表明FaLFY编码区长度为1236bp,编码412个氨基酸,与其他物种的氨基酸序列都有一定的同源性,其中与蔷薇科玫瑰的LEAFY-1同源性最高,达到88%。实时定量RT-PCR结果表明,在不同的组织、不同的花器官中FaLFY的表达量存在差异,其中在花芽中的表达量最高,在营养生长的组织中基本不表达,在成熟花器官中同样不表达,说明该基因在草莓的成花进程中发挥重要作用。
The LFY homologue named FaLFY was isolated from strawberry(Fragaria × ananassa ‘Huaji’)with the method of RT-PCR and RACE. Its expression pattern in different tissues and floral organs was checked with qRT-PCR. The CDS of FaLFY is 1 236 bp in length and it codes a predicted protein of 412 amino acids. The amino acid identity compared with other LFY homologues is highly conserved and shows the highest similarity(88%)with the LEAFY-2 of Rosa canina. The real-time RT-PCR result showed that FaLFY mainly transcripts in reproductive tissues,especially in flower buds, no transcription was detected in vegetative tissues,neither in the four whorls of flower organs. The results showed that FaLFY play an important role in the processing of flowering in strawberry.
