目的 以THP-1来源的巨噬细胞为研究对象,初步研究牙龈卟啉单胞菌(Porphyromonas gingivalis,P.g)感染促进巨噬细胞形成泡沫细胞的影响.方法 以160 nmol/L佛波酯(phorbol 12-myristate 13-acetate,PMA)诱导THP-1单核细胞48 h,使其分化为巨噬细胞.在负荷50 μg/mL低密度脂蛋白的条件下,建立P.g感染巨噬细胞的体外模型,采用胆固醇测定法检测细胞内胆固醇酯含量的变化.结果 经160 nmol/L PMA诱导48 h后,THP-1单核细胞成为巨噬细胞.6h时P.g感染组细胞胆固醇酯(CE)与总胆固醇(TC)比值(CE/TC)为0.445±0.386,24 h时,P.g感染组CE/TC值为0.557±0.430,均显著高于空白对照组和6h组(P<0.05).结论 本研究成功建立了P.g感染巨噬细胞的体外模型,P.g感染可显著促进巨噬细胞总胆固醇和胆固醇酯含量增加,从而可能参与心血管疾病(cardiovascular diseases,CVD)发生和发展.
Objective To investigate the effect of porphyromonas gingivalis (P. g) infection on the formation of foam cell derived from THP-l-derived macrophage. Methods In order to induce macrophage differentiation,THP-1 cells were cultured in the presence of 160 nmol/L PMA for 48 h. And the model of the infection of THP-l-derived macrophage by P. g was established in vitro in the presence of LDL (50 p,g/mL). The amounts of total cholesterol (TC) and cholesterol ester (CE) were quantified using zymochemistry method at 0,6,and 24 h,respectively. Results After the addition of 160 nmol/L phorbol 12-myristate 13-acetate (PMA) for 48 h,THP-1 cells were differentiated into maerophages for further studies. Incubation of LDL-treated macrophages with P. g caused an increase in CE and TC accumulation. Compared with control group,the P. g infection group had significantly increased the ratio of CE to TC (0. 445 ± 0.386) (P〈0.05).Compared with Oh and6h,P, ginfection group had higher ratio (0.557 ±0.430) ofCE toTC (P〈0.05) at 24 b. Conclusions Macropbages can be formed with stimulation PMA to THP-1 monocytes after 48 h; P. g infection can promote macro- phage-derived foam cell formation,which highlights a potential role for P. g in the pathogenesis of cardiovascular diseases ( CVD).
