目的探讨文拉法辛对老年抑郁大鼠海马碱性成纤维细胞生长因子(bFGF)及成纤维细胞生长因子受体1(FGFR-1)表达的影响。方法老年雄性SD大鼠48只,随机分为4组:对照组、模型组、生理盐水组、文拉法辛组,各组12只。模型组、生理盐水组和文拉法辛组均给予单笼饲养加慢性不可预测的温和多相应激方法处理,共5周。文拉法辛组于应激第3周末开始同时给予文拉法辛5mg/(kg·d)治疗,持续14d,生理盐水组同时给予生理盐水14d。对照组不给予任何处理。采用敞箱实验和蔗糖消耗实验评估大鼠抑郁建模情况。采用Westernblot法检测海马bFGF及FGFR1蛋白表达,RTPCR法检测bFGFmRNA及FGFR-1mRNA表达。结果与对照组比较,模型组和生理盐水组海马bFGF、FGFR-1蛋白及mRNA表达均下降,差异有统计学意义(P〈0.01)。与生理盐水组比较,文拉法辛组bFGF、FGFR-1蛋白及mRNA表达均明显增加,差异有统计学意义(P〈0.01)。结论文拉法辛可提高老年抑郁大鼠bFGF、FGFR-1蛋白及mRNA表达。
Objective To study the effect of vanlafaxine on expression of basic fibroblast growth factor(bFGF) and its receptor-1 (FGFR-1) in hippocampus of elderly depressive rats. Methods Forty-eight elderly male SD rats were randomly divided into control group, model group, normal saline treatment group and venlafaxine treatment group(12 in each group). The animals in model group,normal saline treatment group and venlafaxine treatment group were fed in cages with chronic mild stress for 5 weeks. Those in venlafaxine treatment group and normal saline treatment group were given venlafaxine 5 mg/(kg · d)and normal saline respectively for 14 days from the end of week 3. A depressiv test. Expressions of bFGF by Western blot and RT-P protein and mRNA were than in control group and S S e rat model was established by open field and sugar-water consumption and FGFR-1 protein and mRNA in hippocampus of rats were detected CR,respectively. Results The expression levels of bFGF and FGFR-1 ignificantly lower in model group and normal saline treatment group ignificantly higher in venlafaxine treatment group than in normal saline treatment group(P〈0. 01). Conclusion Venlafaxine can increase the expression level of bFGF and FGFR-1 in hippocampus of elderly depressive rats.
