为了优化滑子菇(Pholiota nameko)提取多糖的工艺条件,以滑子菇子实体为原料,用水提醇析法提取粗多糖,在单因素试验基础上,采用3因素3水平的Box-Behnken试验确定提取条件。结果表明,在试验范围内各因素对滑子菇多糖得率的影响依次是提取温度〉料液比〉提取时间;最优工艺参数为料液比1∶46.65(g∶m L),提取温度74.76℃;提取时间4.80 h;最佳提取工艺条件下,响应面拟合所得方程对滑子菇多糖得率预测值为9.12%,实测值为9.05%,实测结果与预测值相差较小,该模型可以较好地对滑子菇多糖提取进行预测。
The extraction conditions of polysaccharides from pholiota nameko was optimized in order to improve extraction efficiency. The pholiota nameko fruiting bodies was used material to extract crude polysaccharide by water and ethyl alcohol. On the basic of single factor test,three-variable and three-level Box-Behnken experimental design were used to optimize process conditions. The results showed that within the tested range of levels, the impact of three process conditions on crude polysaccharide yield decreased in the following order: extraction temperature〉solid-liquid ratio〉extraction time.The optimum technological conditions were found to be solid-liquid ratio was 1:46.65, the best temperature was 74.76℃,the extraction time was 4.80 hours. Under the optimal conditions, the maximum extraction yield of polysaccharides was predicted to be 9.12% and observed to be 9.05%. Therefore, there was a good accordance between the predicted and observed values.
