目的:探讨益气活血方和补肾生髓方对脑缺血再灌注大鼠缺血侧额顶叶皮质Hes-1、Hes-5蛋白表达的动态影响。方法:采用线栓法复制局灶性脑缺血再灌注大鼠模型。随机分为假手术组、模型组、益气活血方组和补肾生髓方组。脑缺血2h,分别再灌注1,2.3周,采用免疫组织化学SABC法检测Hes-1、HeS-5蛋白表达的阳性面积和平均吸收光密度值。结果:在缺血区额项叶皮质。再灌注1-2周时,模型组Hes-1、Hes5表达阳性面积单位、平均吸收光密度值较假手术组明显增加(P〈0.05,或P〈0.01)。再灌注1周时.益气活血方和补肾生髓方组Hes-1、Hes-5蛋白表达阳性面积及平均光密度值显著高于模型组(P〈0.05.或P〈0.01),益气活血方组Hes-1和Hes5蛋白表达平均光密度值显著高于补肾生髓方组(P〈0.01)。再灌注2周时,除补肾生髓方组Hes-5外,益气活血方组和补肾生髓方组Hes-1和Hes5蛋白表达阳性面积及平均光密度值显著高于模型组(P〈0.05,或P〈0.01),益气活血方组Hes-5表达阳性面积显著高于补肾生髓方组(P〈0.01)。再灌注3周时。补肾生髓方组Hes1表达阳性面积,补肾生髓方组和益气活血方组Hes5表达阳性面积及益气活血方组Hes5表达平均光密度值显著高于模型组(P〈0.05)。结论:两种中药复方均能通过增强缺血侧脑组织额顶叶皮质Hes-1和Hes-5的表达,促进局灶性脑缺血后内源性神经干细胞的增殖分化。
Objective: To investigate the dynamic effects of Yiqi Huoxue Recipes (a traditional Chinese prescription for qi-invigorating and blood circulation-activating) and Bushen Shengsui Recipes (a tradition al Chinese prescription for tonifying the kidney to regenerate bone marrow) on the expression of Hes-1 and Hes 5 protein in the ischemic cortex of frontal and parietal lobes in rats with cerebral ischemia-reperfusion injury. Methods: The rat model of local cerebral ischemia-reperfusion was induced by2 hours ofintralumi nal thread occlusion of the middle cerebral artery following after 1-, 2- and 3-week of reperfusion, respec tively. The rats were randomly divided into sham group, model group, Yiqi Huoxue Recipes group and Bushen Shengsui Recipes group. The positive area and mean values of optical density (OD) of Hes 1 and Hes 5 protein expression were observed using the techniques of immunohistochemical staining. Results: The positive area and mean OD values of Hes-1 and Hes 5 protein expression in the ischemic cortex of frontal and parietal lobes in the model group were significantly increased as compared with those in the sham group after 1 and 2-week of reperfusion, respectively (P〈0.05 or P〈0. 01). After reperfusion for 1 week, the positive area and mean OD values of Hes 1 and Hes 5 in the Yiqi Huoxue Recipes group and Bushen Shengsui Recipes group were significantly higher than those in the model group (P〈0.05 or P〈 0.01), and the mean OD values of Hes 1 and Hes 5 protein expression in the Yiqi Huoxue Recipes group were significantly higher than those in the Bushen Shengsui Recipes group (P〈0.01). After reperfusion for 2 weeks, except for Hes 5 protein expression in the Bushen Shengsui Recipes group, the positive area and mean OD values of Hes-1 and Hes 5 in both Yiqi Huoxue Recipes group and Bushen Shengsui Recipes group were significantly increased as compared with those in the model group (P〈0.05 or P〈0.01), and the positive area of Hes 5 protein expicsion in th
