活血利水法对大鼠缺血性脑水肿脑组织内AQP-1和AQP-9表达的影响

目的观察活血利水法对缺血性脑水肿大鼠脑组织内水通道蛋白-1(AQP-1)和水通道蛋白-9(AQP-9)表达变化的影响。方法采用线栓法制作大鼠局灶性脑缺血再灌注模型,随机分为假手术组、模型组、活血利水方低剂量组、活血利水方高剂量组和甘露醇联合川芎嗪组,分别给予相应的药物灌胃;分别于再灌注1、3、7d 3个时间点将大鼠麻醉后采用心脏灌流内固定法取脑,用免疫组化法测定AQP-1和AQP-9表达。结果模型组各时间点AQP-1和AQP-9表达均显著升高。各药物组AQP-1和AQP-9表达均显著降低,1d时甘露醇联合川芎嗪组作用效果最佳,7d时活血利水高剂量组作用效果最佳。结论活血利水法可通过减低脑组织AQP-1和AQP-9的表达来减轻脑水肿程度,抵抗脑缺血再灌注的损伤。

英文摘要:

Objective:To observe the effect of Huoxuelishui Method on AQP-1 and AQP-9 expressions of rats' brain with ischemic cerebral edema.Methods:Ischemia reperfusion rat models made by intraluminal thread occlusion of the middle cerebral artery(MCAO) were randomly divides them into 5 groups as follows:the sham operation group,the model group,the low dose group of Huoxuelishui Decoction,the high dose group of Huoxuelishui Decoction,mannitol and TMP group.After giving them corresponding drugs through gastric perfusion respectively,these expressions including AQP-1 and AQP-9 were detected by the immunohistochemical method.Brain tissues being extracted by the heart reperfusion inner fixation method after 1st,3rd and 7th day anesthesia.Results:The AQP-1 expression and the AQP-9 expression at different time points in the model group were significantly increased.They were significantly reduced in these drug groups.In the 1st day,the effect was the best in the mannitol and TMP group.In the 7th day,there was the optimal result in the the high dose group of Huoxuelishui Decoction.Conclusion:Through reducing the expression of AQP-1 and AQP-9 in brain tissue,Huoxuelishui Method can reduce the brain edema as to resist injury from cerebral ischemia reperfusion.
