背景:建立冠状动脉粥样硬化心脏病(心)阳虚血瘀证大鼠模型,其模型成功率及有效性需进行模型指标体系构建及计算机参数计算后的量化评估。目的:采用计算机软件计算聚类分析统计学方法对温阳活血中药复方影响冠状动脉粥样硬化性心脏病(心)阳虚血瘀证大鼠模型血小板功能及组织形态变化进行分析,以判断模型是否成功建立,温阳活血中药复方是否有疗效。设计、时间及地点:以聚类分析法进行的计算机参数评价,于2004-10/2007-10在辽宁中医药大学动物实验中心进行。材料:清洁级Wistar雄性大鼠60只,将60只大鼠随机分为正常组、模型组、西药对照组、中成药对照组、中药治疗低剂量组、中药治疗高剂量组,10只/组。中药复方由仙茅、桂枝、红参、赤芍、川芎、丹参等组成。方法:正常组大鼠饲喂基础饲料加生理盐水灌胃,其余各组饲喂高脂饲料,模型组大鼠生理盐水灌胃10mL/kg,西药对照组大鼠灌胃阿斯匹林悬液25mg/kg,中成药对照组灌胃复方丹参悬液0.75g/kg,中药治疗低剂量组灌胃中药复方悬液10g/kg(含生药量),中药治疗高剂量组灌胃中药复方悬液20g/kg(含生药量)。主要观察指标:腹主动脉采血检测大鼠心肌酶、血脂及血小板,肾功能各项指标。将60只大鼠42项指标输入电子计算机,应用SPSS13.0统计分析软件进行统计学分析。根据正常组、模型组、温阳活血中药复方治疗组是否自成一类,判断造模方法的特异性和温阳活血中药复方的有效性。结果:温阳活血中药复方能降低造模损伤、降低血小板聚集率、抑制血小板受体活化和信号转导。正常组、模型组、温阳活血中药复方治疗组分别呈现聚集趋势,说明其组内各只大鼠差异性很小。结论:计算参数结果显示,模型建立成功,温阳活血中药复方组能够明显影响冠状动脉粥样硬化心脏病(心
BACKGROUND: To establish rat models of coronary atherosclerotic heart disease (CAHD) due to heart Yang deficiency and blood stasis. Its success rate and efficacy should be assessed by constructing model index system and calculating OBJECTIVE: Using the cluster analysis of the statistical method carries on the analysis to the warming Yang and promoting blood circulation of Chinese traditional medicine compound preparation to platelet's morphous and function on rat model due to heart Yang deficiency and blood stasis of CAHD, to judge the model whether succeeds or not, whether the warming Yang and promoting blood circulation of Chinese traditional medicine compound preparation does have the curative effect. DESIGN, TIME AND SETTING: Cluster analysis was used to evaluate computer reference. Experiments were performed at the Animal Experimental Center of Liaoning University of Traditional Chinese Medicine from October 2004 to October 2007. MATERIALS: Sixty clean Wistar male rats were divided into 6 groups randomly, a normal group, a model group, a western medicine control group, a Chinese formulated products control group, a low dose group of the Chinese materia medica, a high dose group of the Chinese materia medica, 10 in each group.
