目的:研究温阳活血中药复方对冠心病(心)阳虚血瘀证大鼠血小板内受体、信号转导的作用机制。方法:采取高脂饮食、脑垂体后叶素皮下注射及寒冷刺激方法建立冠心病(心)阳虚血瘀证大鼠模型;观察温阳活血中药复方对血小板内受体、信号转导的的影响。结果:温阳活血中药复方组与模型组比较,血浆血管性假血友病因子(vWF)浓度、血栓素B2(TXB2)含量、血小板膜糖蛋白Ⅱb/Ⅲa(GPⅡb/Ⅲa)分子数和α颗粒膜蛋白(GMP-140)含量均明显下降,6-酮-前列腺素F1α(6-keto-PGF1α)含量增高(P〈0.01,P〈0.001)。电镜下模型组血小板腔泡内致密颗粒减少,易于变形,伸出伪足,温阳活血中药复方组血小板表面尚光滑,未见伸出伪足。结论:温阳活血中药复方具有抑制血小板受体的表达、信号转导的作用。
Objective: To research the effect of Wenyang Huoxue Compound Preparation on platelet receptor and signal transduction in coronary heart disease rats with yang deficiency and blood stasis of heart.Methods: The coronary heart disease rat models were established by administering high fat diet,subcutaneous injection with pituitrin and being stimulated by cold methods.Then,to observe the effect of Wenyang Huoxue Compound Preparation on platelet receptor and signal transduction.Results: Compared with model group,the concentration of plasma von Willebrand factor(vWF),the content of thromboxane B2(TXB2),the platelet aggregation rate,the molecular number of GPⅡb/Ⅲa and the content of plateletαgranularmembrane protein(GMP 140) in Wenyang Huoxue Compound Preparation group decreased apparently(P〈0.001),and the content of 6 keto prostaglandin F1α(6-keto-PGF1α) increased significantly(P〈0.001).Under the electron microscope,the dense granules in blood platelet cavity reduced in model group,the blood platelet easy to distort and stretch out the pseudopodium.In Wenyang Huoxue Compound Preparation group,the blood platelet surface was smooth and had not stretches out pseudopodium.Conclusion: Wenyang Huoxue Compound Preparation could constrain the platelet receptor expression and signal transduction.
