目的:观察过氧化物还原酶4(peroxiredoxin 4,Prdx4)蛋白表达的改变对人宫颈癌HeLa细胞迁移和侵袭的影响。方法:采用脂质体瞬时转染法将表达质粒pc DNA3.0-HA-Prdx4转染HeLa细胞;LV-Prdx4 RNAi重组病毒感染HeLa细胞,构建Prdx4 shRNA HeLa细胞系沉默Prdx4的表达;用蛋白质印迹法证实Prdx4蛋白表达水平的变化;应用划痕愈合实验和Transwell迁移及侵袭实验分别检测细胞迁移及侵袭能力的变化。结果:pc DNA3.0-HA-Prdx4质粒转染组He La细胞的Prdx4蛋白表达水平明显上调(P〈0.05),而Prdx4 shRNA HeLa稳定干扰细胞系Prdx4表达水平明显下调(P〈0.05)。Prdx4过表达组HeLa细胞的迁移和侵袭能力明显增强,与空白对照组及空载体转染组比较,差异显著(P〈0.01)。Prdx4表达干扰组HeLa细胞的迁移和侵袭能力明显受到抑制,与空白对照组及阴性对照组比较,差异显著(P〈0.01)。结论:上调Prdx4蛋白表达水平可促进HeLa细胞的迁移和侵袭,在宫颈癌的发生和发展过程中可能起重要作用;下调Prdx4蛋白表达水平可以明显抑制HeLa细胞的迁移和侵袭能力,Prdx4有可能成为临床治疗宫颈癌的一个潜在靶点。
AIM: To investigate the effects of peroxiredoxin 4( Prdx4) protein expression levels on the migration and invasion of human cervical cancer He La cells. METHODS: The plasmid pc DNA3. 0-HA-Prdx4 was transfected into He La cells. The He La cells were infected with LV-Prdx4 RNAi vector to establish stable Prdx4 shRNA He La cells.The change in the expression of Prdx4 protein was validated by Western blotting. The wound-healing assay,and Transwell migration and invasion assays were performed to detect the migration and invasion of He La cells,respectively. RESULTS:The expression of Prdx4 protein was up-regulated in the He La cells after transfection with pc DNA3. 0-HA-Prdx4 plasmid( P 0. 05),whereas it was down-regulated in the Prdx4 shRNA He La cells( P 0. 05). The abilities of migration and invasion were significantly increased in Prdx4-overexpressing He La cells compared with non-transfected and mock plasmid transfected control groups( P 0. 01). When Prdx4 was knocked down by shRNA,the migration and invasion of the He La cells were remarkably repressed compared with blank control group and negative control group( P 0. 01). CONCLUSION:The up-regulation of Prdx4 expression facilitates the migration and invasion of He La cells,and the down-regulation of Prdx4 expression inhibits the migration and invasion of He La cells,indicating that Prdx4 may be a potential molecular target for cervical cancer therapy.
