目的为寻找新的鼻咽癌放疗增敏剂提供实验基础。方法应用MTT法从不同序列的CpGODN中筛选出对经X线照射的人鼻咽癌细胞系CNE-2细胞增殖影响较强的特异序列,观察其对X线照射的CNE-2细胞增殖影响的量效关系及对CNE-2细胞集落形成、细胞迁移的影响。结果CpGODN106可显著抑制经X线照射后CNE-2细胞的增殖,且明显高于单纯照射抑制效应,并呈一定量效关系;CpGODN10630μg/ml能显著抑制CNE-2细胞集落的形成和迁移,且高于单纯照射抑制效应。结论CpGODN106可增强人鼻咽癌细胞CNE-2对X线的放射敏感性,其放射增敏作用可能与其抑制肿瘤细胞的增殖、迁移有关,此为鼻咽癌寻找新的放疗增敏剂提供了实验基础。
Objective To screen the specific sequence CpG ODN from different sequences of CpG ODNs which can enhance the inhibitory effect of X-rays on the proliferation of nasopharyngeal carcinoma cell CNE-2, and observe its impact on tumor radiosensitivity. Methods MTT assay Was used to select the stronger specific sequence from different sequences of CpG ODNs screened by the X-ray irradiation,which have an impact on the proliferation of nasoplmryngeal carcinoma cell line CNE-2 cell, and observe the dose-effect relationship between CpG ODN106 and proliferation of CNE-2 cells treated by X -ray irradiation. Colony-forming tests were used to observe the effect of CpG ODN106 on the colony-forming and migration of CNE-2 cells treated by X-ray irradiation. Results After X-ray irradiation, CpG ODN106 eouM significantly inhibit CNE-2 cell proliferation. It showed some dose-effect relationship;CpG ODNI106 significantly inhibited the formation and mi- gration of CNE-2 cell colony at a concentration of 30 μg/ml compared with the radiation alone, and the inhibitory effect of irradiation was increased significantly. Conclusion CpG ODN106 could enhance radiation sensitivity of human nasopharyngeal carcinoma cell CNE-2 response to X-rays.
