目的通过观察β-catenin和cox-2在体外培养的毛囊隆突区细胞中的表达,探讨其对隆突区细胞增殖的作用及相互关系。方法在体外分离培养毛囊隆突区细胞的基础上,采用免疫细胞化学方法分别检测不同时相β-catenin和cox-2在毛囊隆突区细胞中的表达。结果β-catenin和cox-2在毛囊隆突区细胞呈强表达,并表现出与培养时间的相关性;同时β-catenin可见于细胞质与细胞核中,而cox-2仅见于细胞核。结论β-catenin和cox-2对毛囊隆突区细胞具有促增殖作用,cox-2可能是β-catenin途径发挥作用的核内靶基因。
Objective To study the expression of and investigate relationship of their expression on ce β-catenin and cox-2 in the bulge cells of hair follicle 11 proliferation. Methods The hair follicle was prepared from the resected cheek skin of 20 Wistar rats aged 7 days. The bulge cells were resected from the intact hair follicle and cultured in vitro. Immunocytochemical technique ( ICC ) was applied to detect β-catenin and cox-2 expression in bulge cells at culture day 3, 5, 8, 13. Results β-catenin and cox-2 strongly expressed in bulge while cells and the expression correlated with the culture days. β-catenin appeared in both plasma and nucleus, cox-2 only in nucleus. Conclusion β-catenin and cox-2 were correlated with bulge cells proliferation, and cox-2 might be the target gene of β-catenin signaling pathway in nucleus.
