目的研究二十味沉香丸降低高原性肺高动脉高压的机制。方法将200只Wistar大鼠随机分成平原组、高原对照组和给药组,高原对照组和给药组在第1、3、7、15、30天分别取20只进行测定。采用MP150生理信号采集系统、ELISA及WB等方法测定各组大鼠的肺动脉压(PAP)、血清HIF-1α和CaMKⅡδ水平。结果高原对照组大鼠的PAP随在高原居住时间的延长而升高,7 d后明显升高,给药组的升高不明显;高原对照组与平原组、给药组相比有显著差异性(P〈0.01);给药组与平原组相比无明显差异;与平原组的比较,给药组大鼠的血清HIF-1α升高不明显,高原组的显著升高(P〈0.001);与给药组的比较,高原对照组的CaMKⅡδ水平在急性期显著升高(P〈0.05),但后期有所下降,而给药组的升高不明显,但第30天时的水平接近高原对照组。结论二十味沉香丸在早期低氧期可明显通过降低HIF-1α和CaMKⅡδ水平降低大鼠的PAP,但第30天时的作用不明显,表明沉香丸预防急性低氧所致PAP升高的作用优于慢性低氧。
OBJECTIVE To study the mechanism of 20Wei Chenxiang pills reducing pulmonary artery pressure on the plateau. METHODS 200 plain Wistar rats were randomly divided into plain group, the high altitude control group and the high altitude drug treatment group. 20 rats from the control group and drug group respectively were measured on the 1 st,3rd,7th, 15th and 30th day. The PAP, HIF - 1 α and CaMK Ⅱδ level were measured or determined by MP150 physiological signal acquisition system, ELISA and WB. RESULTS The PAP of rats were increased in the high altitude control group with the extension of the plateau residence time, espe- cially after 7 days, significantly increased, but no obvious increased in the drug group. It is markedly different between high altitude control group and plain/high altitude drug treatment group (P 〈 0.01 ). Drug treatment group, compared with the plain group, showed no significant difference. Compared with the plain group, serum levels of HIF - 1 α in the drug treatment group was not increased obvi- ously, and serum levels of HIF - 1 α levels in the high altitude control group were significantly increased ( P 〈 0.01 ) compared with the drug treatment group. CaMK Ⅱδ levels in the high altitude control group were increased significantly in acute hypoxic phase (P 〈 0. 05), but later declined in the drug treatment group was not increased obvious, but at 30th day the level closed to that of the high altitude control group. CONCLUSION Tibetan drugs ,20Wei Chenxiang pills,could significantly reduce PAP by decreasing HIF - 1α and CaMK Ⅱδ level during the early hypoxia, but the reductive effect at 30th day is not significant. It also shows that effect of 20Wei Chenxiang pills in acute hypoxia is superior to chronic hypoxia on prevention of rising of PAP.
