目的:从microRNAs角度揭示HBeAg(-)轻度慢乙型脾胃湿热证发生的分子机制。方法:将符合标准的病例分为HBeAg(-)轻度慢乙肝脾胃湿热证组与HBeAg(-)轻度慢乙肝无症状组,借助AgilentHumanmiRNA8×60k微阵列芯片检测血浆中microRNAs表达谱,求得两组间的差异表达microRNAs谱(P〈0.05),借助miRNA生物信息学分析软件预测其靶基因并对靶基因选行GO功能富集分析和pathway分析。结果:两组间的差异表达microRNAs共86条(P〈0.05),37条上调,49条下调;GO分级及Pathway分析得到其功能主要涉及细胞黏附、生物合成过程的正/负调控、蛋白质定位、Wm信号通路、RNA的代谢过程.正/负调控基因表达的正/负调控、蛋白氨基酸的磷酸化、Notch信号传导途径、细胞凋亡、Hedgehog信号通路、T细胞受体信号通路等。结论:HBeAg(-)轻度慢乙肝脾胃湿热证受特异性差异mieroR-NAs调控,并涉及多个生命过程,其中Wnt信号通路可能是HBeAg(-)轻度慢乙肝脾胃湿热证发生的关键分子机制。
Objective:From microRNAs to discuss the expression and molecular mechanisms between spleen - stomach damp -heat syndrome and compliant asymptomatic group of mild chronic hepatitis B with positive HBsAg, HBeAg and HBcAb. Methods :The patients were divided into spleen -stomach damp -heat syndrome group and compliant asymptomatic group of mild chronic hepatitis B with positive HBsAg, HBeAg and HBcAb. The fasting serum samples were collected and the expression profile of microRNAs in plasma was detected using Agilent Human miRNA 8 - 60 k microarrays. The data was analyzed using SPSS19. 0 software and online SAS system to obtain the differences expressed microRNAs spectrum between the groups (P 〈 0. 05). Through the means of bioinformatics analysis software, analyze and predict the target genes with GO enrichment target genes KEGG pathway analysis. Results:The difference between the two types was totally 86 microRNAs expression (P 〈 0. 05 ), 37 up- regulated,49 down- regulated. Analyze target genes GO enrichment and suggest regulating target genes and their participa- tion of cell adhesion, biosynthesis of positive/negative control, protein localization, Wnt receptor signaling pathways, RNA metabol- ic processes of positive/negative regulation of gene expression in the positive/negative control, protein amino acid phosphorylation and other vital processes. Pathway analysis results were mainly involved in Notch signaling pathway, apoptosis, Wnt signaling path- way, Hedgehog signaling pathway and T cell receptor signaling pathways. Conclusion: There are multiple differences of microRNAs between spleen - stomach damp - heat syndrome group and compliant asymptomatic group of mild chronic hepatitis B with positive HBsAg, HBeAg and HBcAb, which regulate more than one life processes and the mechanisms, importantly, Wnt signaling pathway may be a key molecular mechanism in spleen - stomach damp - heat syndrome group of mild chronic hepatitis B with positive HB- sAg, HBeAg and HBeAb.
