目的 研究发酵液中紫杉醇的萃取工艺.方法 通过高效液相色谱含量测定法比较了多种有机溶剂萃取紫杉醇的效果.结果 将发酵液过滤得菌丝反复冻融在研钵中研碎,按菌丝重量和加入甲醇体积1:10的比例(g:mL)进行甲醇搅拌提取,过滤后得甲醇提取清液;再将甲醇提取清液与发酵液滤去菌丝后得清液合并,合并液中加入1倍体积二氯甲烷,每次萃取4 h、萃取3次,紫杉醇萃取率最高.结论 筛选出了发酵液中紫杉醇的最佳萃取工艺.
Objective To investigation of extraction process of taxol in fermentation broth. Methods A variety of organic solvents was compared in extraction process of taxol. The content of taxol was determinated by I-IPLC. Results The taxol extraction amount was the maximum when added one time volume dichloromethane in the mixed liquid of clear illtrate without hyphae and methanol extraction liquid from the process that hyphae triturated after repeated freeze thawing was extracted thrice by methanol in the ratio of one to ten (g: mL), extracted for four hours each time. Conclusion The optimal extraction process of taxol in fermentation broth was screened out.
