为探讨一种新型的神经营养因子,Neuritin在神经系统成熟及其损伤后的再生修复过程中发挥着重要的调节作用。为了获得大鼠Neuritin基因转录本及其蛋白结构的基础数据,本研究利用比较基因组学分别设计与小鼠2个转录本同源的引物,采用RT—PCR的方法钓取大鼠Neuritin基因。采用生物信息的方法对大鼠Neuritin的序列同源性及蛋白结构进行全方位分析。结果表明,大鼠只存在一个Neuritin转录本,编码142个氨基酸,与其他动物的编码序列高度同源;三级蛋白结构分析结果显示大鼠Neuritin蛋白富含有a螺旋,其结构和理化性质无异于人类Neuritin基因。由此可知,本研究首次厘定了大鼠NeuritinmRNA,其蛋白质结构的深入分析也为进一步理解大鼠Neuritin基因的生理功能奠定了基础数据。
As a new neurotrophic factor, Neuritin plays an important regulatory role in the mature nervous system and regeneration process.To obtain fundermental data for rat Neuritin transcript and its protein structure,comparative approaches were employed to design two primers which were homologous to the two transcripts of mouse Neuritin,and RT-PCR method was used to fish rat Neuritin transcripts.And bioinformatics methods were employed to analyze rat Neuritin sequence homology and its protein structure.The studies indicated that there was only one Neuritin transcript in rat,encoding 142 amino acids,which was highly conserved among species;Tertiary structure analysis showed that rat Neuritin was made up of six (x-helix,and its structure and physicochemical properties was tantamount to the human Neuritin. In this paper, rat Neuritin transcript was firstly defined,and comprehensive analysis of rat Neuritin structure layed the foundation data for further understanding of its physiological functions.
