MIN6细胞给予400 μmol/L H2O2处理分为正常对照组、正常加H2 O2组、Scramble siRNA(Small interfering RNA)组和RIP140 siRNA组,400 μmol/L H2O2能增加MIN6细胞的凋亡率(27.89%±1.91%对3.74%±0.55%,P<0.01)和丙二醛含量[(35.17±6.26对20.07±1.86) nmol/mg,P<0.05],明显减少MIN6细胞活力(24和48 h,均P<0.01)、总超氧化合物歧化酶水平[SOD,(4.79±0.85对3.09±0.60) U/mg,P<0.05].下调受体相互作用蛋白140(RIP140)表达能减少H2O2诱导的MIN6细胞凋亡率(11.45% ±2.27%对30.39%±5.32%,P<0.01)和丙二醛含量[(22.32±1.94对33.42±2.51)nmol/mg,P<0.01],增加总SOD水平[(5.32±0.40对3.09 ±0.15)nmol/mg,P<0.01)].
MIN6 cells cultured with 400 μmol/L H2O2 were divided into normal control group,control group with 400 μmol/L H2O2,scrambled siRNA group,and RIP140 siRNA group.400 μmol/L H2O2 could significantly increase apoptosis rate of MIN6 cells(27.89% ± 1.91% vs 3.74% ±0.55%,P<0.01) and the content of maleic dialdehyde [M DA,(35.17 ± 6.26 vs 20.07 ± 1.86) nmol/mg,P<0.05],suppress cell viability (24 and 48 h,both P<0.01),and lower the level of total superoxide dismutase [SOD,(4.79 ± 0.85 vs 3.09 ± 0.60) U/mg,P< 0.05].When receptor interaction protein 140 (RIP140) expression was suppressed by RNA interference,the apoptosis rate of MIN6 cells (11.45 % ± 2.27% vs 30.39% ± 5.32%,P<0.01) and the content of MDA [(22.32 ± 1.94 vs 33.42 ± 2.51) nmol/mg,P<0.01] were reduced,and levels of total SOD was increased [(5.32 ± 0.40 vs 3.09±0.15) nmol/mg,P<0.01)].
