目的:研究梓醇(Catalpol)对高糖高脂诱导的人脐静脉血内皮细胞(HUVECs)炎症反应的影响。方法:体外培养人脐静脉血内皮细胞,用CCK-8法检测不同浓度梓醇溶液对HUVECs细胞增殖活性的影响。将细胞分为空白对照组、高糖高脂组(给予33mmol/L葡萄糖+500μmol/L棕榈酸作用48h)、高糖高脂+1 000mg/L梓醇预处理组(梓醇1 000mg/L预处理24h后,给予33mmol/L葡萄糖+500μmol/L棕榈酸作用48h,以下简称梓醇预处理组),收集细胞上清液,用ELISA检测上清液中的炎症因子TNF-α、IL-6的浓度。结果:梓醇作用于HUVECs细胞的最佳浓度为1 000mg/L。高糖高脂组中TNF-α和IL-6浓度较空白对照组均明显增高(t=7.03、6.68,P均〈0.01)。与高糖高脂组比较,梓醇预处理组上清液中炎症因子TNF-α及IL-6浓度均下降(t=-2.87、-3.76,P均〈0.05)。结论:高糖高脂可诱导血管内皮细胞炎症反应,而梓醇对高糖高脂诱导的炎症反应有一定的改善作用。
Objective:To investigate the effects of catalpol on the inflammatory factors of human umbilical vein endothelial cells(HUVECs)induced by high sugar and high lipid.Methods:HUVECs were cultured in vitro.CCK-8method was used to test cell viability.And then the cells were divided into 3groups:control group,high glucose and high fat group(33mmol/L glucose and 500μmol/L palmitic acid,48h)and1 000mg/L catalpol group.The consistences of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in the supernatant were detected by enzyme-linked immunosorbent assay(ELISA)to observe the inflammation reaction.Results:The consistences of TNF-αand IL-6in high sugar and high lipid group were obviously higher than those in control group(t=7.03,t=6.68,P〈0.01).The consistences of TNF-α(t=-2.87,P〈0.05)and IL-6(t=-3.76,P〈0.05)in 1 000mg/L catalpol group were lower than those in high sugar and high lipid group.Conclusion:High sugar and high lipid can induce inflammatory reaction.Catalpol,in some degree,inhibits the inflammation of HUVECs induced by high sugar and high lipid.
