为研究FnBPA-A不同遗传多态性重组质粒抗血清对牛源金黄色葡萄球菌与牛原代乳腺上皮细胞粘附作用的影响。采集奶牛乳腺组织样品,用组织块种植法和胰酶消化法进行纯化和传代培养,并利用对乳腺上皮细胞特异性角蛋白18进行鉴定。将4株不同金黄色葡萄球菌分离株作用于纯化的乳腺上皮细胞,分析不同菌株粘附牛乳腺上皮细胞的能力。进而比较研究FnBPA-A不同遗传多态性重组质粒免疫后的抗血清对4株不同金黄色葡萄球菌分离株与牛乳腺上皮细胞粘附的抑制作用。结果表明,分离纯化获得了牛原代乳腺上皮细胞,分析了该乳腺上皮细胞的培养特性,不同金黄色葡萄球菌分离株对该细胞粘附的检测结果表明,4株不同分离株粘附牛乳腺上皮细胞的能力明显不同(P〈0.05),其中GY278菌株粘附乳腺细胞的能力最强。FnBPA-A不同遗传多态重组质粒免疫后的抗血清抑制不同菌株粘附牛乳腺上皮细胞的能力也表现出明显的差异性,pV-819、pV-309、pV-20-2免疫组的抗血清对4株牛源金黄色葡萄球菌分离株的抑制作用高于其他免疫组。
In order to study the effects of different FnBPA-A genetic polymorphism recombinant plasmid anti-serum on adhesions between bovine Staphylococcus aureus and bovine mammary epithelial cell.The bovine mammary tissue samples,tissue explants method and trypsinization method were used to conduct purification and hand from generation to generation,which were determined by cytokeratin 18 of bovine mammary epithelial specificity.The 4different S.aureus isolates acted on purification mammary epithelial cell and analyzed the capacity of different strains sticking on the bovine mammary epithelial cell,and then the inhibition effect of anti-serum of different FnBPA-A genetic polymorphism recombinant plasmid on 4different S.aureus isolates acting on mammary epithelial cell were investigated.The result showed that the bovine mammary epithelial cell were built by distribution purification and cultivation characteristics of mammary epithelial cell were analyzed.The effects of 4different S.aureus isolates on sticking on bovine mammary epithelial cell showed that the capacity of 4different isolates sticking on bovine mammary epithelial cell were significantly different(P〈0.05),among which GY278 had the strongest capacity sticking on bovine mammary epithelial cell.The capacity of anti-serum inhibiting different strains sticking on bovine mammary epithelial cell of FnBPA-A different genetic polymorphism recombinant plasmid also indicated significant differences.The inhibition capacity of pV-819、pV-309、pV-20-2immune group anti-serum were higher than that of other experimental groups.
