目的:观察伤寒沙门菌鞭毛转子蛋白基因fliG在H:z66抗体应激后对其他基因表达的影响。方法:利用自杀质粒介导的同源重组法制备伤寒沙门菌fliG基因缺陷变异株;利用伤寒沙门菌全基因组芯片分析技术,比较伤寒沙门菌野生株和fliG基因缺陷变异株在H:z66抗体应激后的基因表达谱差异,并选择部分表达有差异的基因进行实时定量PCR验证。结果:经PCR验证及序列比对,伤寒沙门菌fliG基因缺陷变异株制备成功;基因表达谱比较结果表明,与野生株相比,伤寒沙门菌fliG基因缺陷变异株在H:z66抗体应激后有21个基因表达上调,7个基因表达下调;实时定量PCR结果与芯片结果一致。结论:fliG基因在伤寒沙门菌受到H:z66抗体刺激后的基因表达调控中发挥一定作用。
Objective: To explore the influence of flagella rotor protein gene fliG on gene expression regulation of Salmonella enterica serovar Typhi under anti-z66 antiserum stress.Methods: The fliG deleted mutant of S.enterica serovar Typhi was prepared by homologous recombination mediated by suicide plasmid;S.enterica serovar Typhi genomic microarray was used to investigate the difference of gene expression between the wild type and fliG mutant;qRT-PCR was done to prove the results of microarray.Results: The fliG deleted mutant of S.enterica serovar Typhi was constructed successfully.Gene expression profiles analysis revealed that expression of 21 genes were increased and expression of 7 genes were decreased in the fliG mutant under anti-z66 antiserum stress.Conclusion: The flagella rotor protein gene fliG probably played a role to regulate some gene expression in response to anti-z66 antiserum stress.
