以发育中的小麦籽粒为材料,对两种淀粉分支酶凝胶电泳活性染色方法的缓冲体系、染色过程、染色时间,试剂浓度等进行了比较验证。结果表明,Nagamine等文献中的酶活性染色方法能很好的区分淀粉分支酶的同种型类型和基因位点分布,显示不同基因位点的遗传多态性特点,与Yamanouchi等文献中的染色方法相比,酶带分辨率高,电泳图谱清晰,实验成本降低,可为淀粉分支酶的分子生物学及遗传特性研究提供一种经济、可靠的方法。
The present two enzyme activity staining of starch branching enzyme(SBE)using native polyacrylamide gel electrophoresis(Native-PAGE) was compared with developing wheat grain in several aspects,such as buffer system,staining process,staining time as well as chemicals concentration in the experiment.The result showed that Nagamine's method could identify isoform types and gene loci of SBE,which exhibited genetic polymorphism for different gene locus.In comparison with Yamanouchi's experiment method,the Nagamine's possessed higher differentiation of zymograms,better definition of stained electrophoretogram and lower experimental expenses,which would provide an authentic experimental means for research of molecular biology and genetics on SBE.
