利用稀释涂布法对芦笋老茎堆肥不同发酵阶段6个样品中的嗜热细菌进行分离,并采用16S rDNA序列分析方法对分离得到的菌落形态有明显区别的22株细菌进行鉴定。根据16S rDNA序列分析结果,22株细菌菌株中13株属于芽胞杆菌属(Bacillus),1株属于类芽胞杆菌属(Paenibacillus),4株属于假黄色单胞菌属(Pseudoxanthomonas),1株属于肠杆菌属(Enterobacter),1株属于副球菌属(Paracoccus),1株属于短芽胞杆菌属(BrevibaciUus),菌株D小2在GenBank数据库中未找到与其相似的已知细菌属的序列,分类地位待定。从以上鉴定结果可以看出,芦笋老茎堆肥中的优势嗜热细菌主要是芽胞杆菌(Bacillusspp.)和假黄色单胞菌(Pseudoxanthomonasspp。)。
Thermophilic bacterial strains in six asparagus bld stem compost (AOSC) samples at different fermentation stages were isolated using dilution-spreading-plate method, and twenty-two bacterial strains were identified according to their apparent morphological distinction using 16S rDNA sequence analyses. According to 16S rDNA sequence analysis results thirteen of 22 strains were identified as Bacillus spp. , one as Paenibacillus sp. four as Pseudoxanthomonas spp. , one as Enterobacter sp. , one strain was identified as Paracoccus sp. , and one as Brevibacillus sp. Strain D-b2 could not be identified due to no similar known bacterial sequence to its sequence was searched in the GenBank, there- fore, its classification position was still remained to be confirmed. From the characterization results above, one could know for that the dominant thermophilic bacteria in the AOSC were mainly Bacillus spp. and Pseudoxanthomonas spp.
