目的:研究SV40 Large T(LT)对淋巴细胞转化的影响。方法:将逆转录病毒载体质粒pBabe-SV40LT转入外周血单核细胞。倒置显微镜观察细胞形态变化,蛋白印迹明确LT蛋白表达,流式细胞仪分析膜表面分子的表达,G显代染色体确定核型,生长曲线判别细胞生长状态。软琼脂实验检测细胞恶性变。结果:导入SV40LT后的淋巴细胞形态发生明显改变,由悬浮生长变为贴壁生长。转染细胞中可以检测到LT抗原的表达,有67%细胞表达CD56+分子,核型为异倍体,可在软琼脂上形成细胞集落。结论:用SV40LT转染获得一株可稳定传代且具有CD56+的异倍体恶性淋巴细胞。
Objective To study the effects of SV40 large T on human-derived lymphocyte transformation.Methods Peripheral blood mononucleate lymphocytes were conducted with the retroviral vector plasmid pBabe-SV40 large T.The cells morphological changes were watched by inverted microscope.The protein of large T antigen was detected by Western blot.Lymphocyte surface molecules were identified by flow cytometry.The cell karyotype was analyzed by G-banding.The cell proliferation was described by growth curve.The soft agar assay was performed to examine the cells malignant transformation.Results Some significant morphological changes were observed in the SV40LT gene transfected human-derived lymphocytes is accompanied by transition from suspension to adherent growth.The expression of large T antigen was found in the transfected cells.Moreover,67% cells showed CD56 marker and were able to grow in soft agar.Conclusion SV40 large T antigen successfully induced a stable passaging and malignant transformation of lymphocyte with expression of CD56+.
