目的探讨胆碱能受体(muscarinic acetylcholine receptors,M受体)各亚型在大鼠膀胱ICC样细胞(interstitial cells of Cajal,ICC)的表达及意义。方法制备SD大鼠膀胱组织铺片,通过免疫荧法检测大鼠膀胱组织内ICC样细胞分布情况,并进一步采用免疫荧光双标法(c-kit抗体及M受体亚型抗体)检测膀胱ICC样细胞上M受体亚型的表达。结果在大鼠膀胱肌层及肌间发现膀胱ICC样细胞呈散在或者彼此连接的分布,M受体的5种亚型中,ICC样细胞上不表达M1、M4、M5受体亚型,表达M2和M3两种受体亚型。结论胆碱能神经可能通过激活膀胱ICC样细胞上胆碱能受体来调节膀胱功能。
Objective To investigate the expressions of subtypes of muscarinic acetycholine receptors and to explore the roles of the receptors on interstitial cells of Cajal-like ceils of SD rat bladder. Methods Stretched preparations of SD rat bladder were made firstly. Then immunohistochemical analyses with c-Kit as markers for ICC-like cells were performed. Immunoflurorescence (double-label) was used to detect the expres- sion of subtypes of the receptors in ICC-like cells. Results C-kit positive cells in SD rat bladder were mainly distributed at the edge of smooth muscle bundles and among smooth muscle scattered or conjoining in network. We found that there were only M2 and M3receptor subtypes but no M1, M4 or M5 on ICC-like cells of SD rat bladder. Conclusion ICC-like cells may be activated by cholinergic nerve via cholinergic receptors on them to modulate the bladder function.
