目的:探讨孕18周胎儿膀胱是否有ICC样细胞的分布,为人膀胱ICC样细胞发育研究提供依据。方法:制备孕18周胎儿膀胱组织冰冻切片,通过c-kit多克隆抗体和平滑肌肌动蛋白单克隆抗体免疫荧光双标,激光共聚焦显微镜观察检测胎儿膀胱组织内ICC样细胞分布情况。结果:孕18周胎儿膀胱组织发现具有纺锤形的细胞体、椭圆形细胞核和突触等典型特征的ICC样细胞分布,位于平滑肌肌柬边缘和肌柬间,散在分布,未连接成网络。结论:首次报道人膀胱ICC样细胞在孕18周胎儿时已经发育。
Objective: To detect whether there are interstitial cell of Cajal (ICC)-like cells in bladder of 18-week-old foetus, which may be a basement for the further study on the development of ICC-like cells in human bladder. Methods: Cryosections of bladder tissue of a 18-week-old foetus were labeled with primary(polyclonal antibody of C-kit (Anti-c-kit))and fluorescent secondary(monoclon- al antibody of smooth muscle actin (anti-SMA))antibodies, and imaged using confocal laser scanning microscopy to examined the distri- bution of ICC-like cells in the bladder tissue. Result: We found that there are ICC-like cells, which have atractoid bodies,ellipsoidal nucleus and synapses, in lhe bladder tissue of the 1 g-week-old foetus. It was most often seen at the edge of smooth muscle bundles and between smooth muscle bundles. They distribute independently without forming a network. Conclusion: We found the ICC-like cells in human bladder have developed in foetus as early as 18 Weeks old at first time.
