[目的]研究蓝莓花青素(BA)对人结肠癌Lovo细胞增殖及凋亡的影响,分析凋亡相关基因—p53基因mRNA和蛋白表达的变化。[方法]分别用浓度为50μg/ml、100μg/ml、200μg/ml的BA处理结肠癌Lovo细胞24h,采用MTT法、免疫荧光法和Real-time PCR法和免疫细胞组织化学分析BA对Lovo细胞增殖、细胞凋亡、p53 mRNA和蛋白表达的影响。[结果]BA呈剂量依赖的方式抑制Lovo细胞增殖;能诱导细胞发生晚期凋亡;与对照组相比,不同浓度BA处理后p53基因mRNA表达水平分别上调0.602、0.697、0.541倍(P〈0.05),但BA不同剂量组间并无明显的差异(P〉0.05);免疫细胞组化结果表明随着BA浓度的增加,p53表达增强。[结论]BA可以诱导结肠癌Lovo细胞发生晚期凋亡,p53基因mRNA表达上调,但与BA的浓度无相关性。
[ Objective] Investigate the effects of blueberry anthocyanidins(BA) on apoptosis in colon cancer Lovo cell,and analyze mRNA expression of apoptosis - related gene of p53. [ Methods ] After colon cancer Lovo cell were treated with different concentrations of BA (50gμg/ml, 100μg/ml,200μg/ml) for 24 hours, apoptosis and mRNA expression of p53 were analyzed using MTT,Annexin V -FITC/PI double staining method,Real- time quantitative polymerase chain reaction( RT- PCR) and Immunocytochemistry analysis. [ Results] An- nexin V - FITC/PI double staining experimental result shows that rate of apoptotic was risen with the increasing concentration of BA. The results of RT - PCR show that mRNA expression of p53 gene in BA treated group was increased O. 602,0. 697,0. 541 times compared with control group( P 〈 0. 05 ), but it was no difference in different dose groups. Immunocytochemistry analysis showed that the expression of p53 was obviously increased after the BA treatment. [ Conclusion] BA can induce cell occurred late apoptosis and mRNA expression of p53 genes increase which was not correlated with the concentration of BA.
