目的观察蓝莓花青素对结肠癌LS174T细胞增殖的抑制作用及凋亡的诱导及探究蓝莓花青素的抗癌机制。方法体外培养结肠癌LS174T细胞,加入不同浓度的蓝莓花青素(25、50、100和200μg/m L)处理,利用CCK法、Hoechst染色检测不同浓度的蓝莓花青素对LS174T细胞增殖和凋亡的影响;实时荧光定量PCR(real-time PCR)检测细胞相关凋亡基因P53、CCND3、P73、SFN、caspase-8和FAM200a mRNA的表达;蛋白质免疫印迹法(Western blot)检测细胞caspase-8和P73的表达。结果蓝莓花青素呈时间和剂量依赖性抑制了结肠癌LS174T的增殖(P〈0.05);蓝莓花青素呈现剂量依赖性诱导癌细胞凋亡;CCND3、P73和SFN mRNA表达水平上调(P〈0.05),caspase-8、FAM200a和P53 mRNA表达水平下调(P〈0.05);caspase-8蛋白表达下调,P73蛋白表达上调。结论蓝莓花青素抑制结肠癌ls174t细胞增殖,诱导凋亡与P73蛋白表达上调和caspase-8蛋白表达下调相关。
Objective To observe actions and effects of anthocyanins on antiproliferation and proapoptosis of human colon cancer cells LS174 T,investigate anti-cancer mechanism of blueberry anthocyanin. Methods Colon cancer cells LS174 T were cultured in vitro,treating LS174 T cells with blueberry anthocyanins( 25,50,100 and200 μg / m L). We tested the effects of blueberry anthocyanins on proliferation and apoptosis by CCK and Hoechst33258 chromatin staining dye; mRNA expression of apoptosis-related genes( including P53,CCND3,P73,SFN,caspase-8 and FAM200 a) was measured by real-time PCR. In addition,protein level of caspase-8 and P73 was also analyzed using Western blot. Results The blueberry anthocyanins suppress the proliferation and induce apoptosis of LS174 T cells in a dose-dependent and time-dependent manners; the expression of CCND3,P73 and SFN mRNA are up-regulated while the expression of caspase-8,FAM200 a and P53 mRNA are down-regulated;the expression of protein levels of caspase-8 and P73 are down-regulated and up-regulated. Conclusions Blueberry anthocyanins induce cell cycle arrest and apoptosis of colon cancer LS174 T cells,which is associated with up-regulation of the expression of tumor suppressor P73 and a down-regulation of the expression of caspase-8.
