目的:通过定量检测和比较Attractin(Atrn)mRNA和Atrn蛋白在小鼠睾丸发育过程中的动态变化,为进一步研究Atrn在睾丸的生理作用提供实验依据。方法:用相对定量法检测Atrn mRNA在生后1、5、10、15、21、28、35、42、56和120天龄小鼠睾丸发育过程中的表达变化,并用Western blot方法检测各天龄小鼠睾丸Atrn蛋白表达。结果:Atrn mRNA和Atrn蛋白在小鼠睾丸发育过程中的表达变化趋势相似,在生后第1天两者均有表达,但是处于相对较低水平,此后是一个逐渐增高的过程,在28天出现较明显上调。在这之后,两者表达变化出现差异。Atrn mRNA在生后28~35天表达最丰富,随后稳定于这一水平,Atrn蛋白表达则是继续增加直至成年。结论:Atrn参与精子发生的启动、精原干细胞的增殖分化、精母细胞的减数分裂和精子形成等过程,并有助于Leydig细胞进一步成熟。
Objectives:To study the role of Attractin(Atrn)in mouse testis,mRNA and protein expression of Atrn in mouse testis during post-natal development were investigated.Methods:The mRNA and protein expression of Atrn in mouse testis during post-natal development(at day 1,5,10,15,21,28,35,42,56and120postpartum)were investigated by real time RT-PCR and Western blot separately.Results:The mRNA and protein expression of Atrn had similar tendency in mouse testis during post-natal development.Relative low level of Atrn mRNA and protein were detected in mouse testis on day 1,then increased obviously at day 28.Expression of mRNA remained this level afterwards,but protein expression increased gradually as the animal aged till maximal level in the adult testis.Conclusion:It was presumed that Atrn may be involved in spermatogenesis and Leydig cell development.
