目的:探讨敲除Attractin(Atrn)或Mahoganoid(Md)基因对雄性小鼠生殖内分泌激素的影响。方法:选取成年雄性Atrn基因敲除纯合子、Md基因敲除纯合子及正常小鼠(均为C3H/Hej种系)各12只设为Atrn基因敲除组、Md基因敲除组和对照组。内眦静脉取血清取外周血,化学发光法检测血清睾酮(T)、雌二醇(E2)浓度,用酶联免疫法(ELISA)检测血清卵泡刺激素(FSH)、黄体生成素(LH)和泌乳素(PRL)浓度,进行组间浓度对比。结果:Atrn基因敲除组和Md基因敲除组的外周血E2、LH、FSH、PRL浓度均低于对照组,差异有统计学意义(P〈0.05);血清T浓度Atrn基因敲除组高于对照组,而Md基因敲除组低于对照组,差异无统计学意义(P〉0.05)。结论:Atrn或Md基因敲除后雄性小鼠的外周血E2、LH、FSH、PRL浓度均有下降,其具体机制及对外周血T浓度的影响有待进一步研究。
Objective: To probe into the influence of the level of reproductive endocrine hormone of male mice after Attractin(Atrn) or Mahoganoid(Md) gene knockout. Methods: Male mature Atrn gene knockout homozygote mice, Md gene knockout homozygote mice and wild type mice(C3H/hej) were divided into 3 groups: Atrn knockout group, Md knockout group and control group. Each group had 12 mice. Blood samples were collected from angular vein of mice. The levels of serum T and E2 were determined with CLIA. The levels of serum FSH, LH and PRL were determined with ELISA. Results: The levels of serum E2, FSH, LH and PRL of Atrn knockout group and Md knockout group were lower than those in control group(P〈0.05). The serum T level of Atrn knockout group was higher than that in control group(P〉0.05). The serum T level of Md knockout group was lower than that in control group(P〉0.05). Conclusion: The levels of serum E2, FSH, LH and PRL of male mice are down-regulated after Atrn or Md gene knockout. Its mechanism and whether the serum T level is affected need to be studied further.
