目的:探讨Attractin(Atrn)在培养大鼠睾丸leydig细胞的表达。方法:体外培养成年雄性sD大鼠睾丸leydig细胞,采用实时荧光定量RT-PCR检测Atrn mRNA表达,western blot检测Atrn蛋白的表达,同时应用化学发光法检测细胞睾酮分泌水平。结果:在体外培养大鼠睾丸leydig细胞过程中,Atrn mRNA、Atrn蛋白表达量和睾酮生成量逐渐减少。Atrn蛋白表达量与Atrn mRNA表达量(r=0.953,P〈0.05)和睾酮生成量(r=0.976,P〈0.01)成正相关。结论:大鼠睾丸Leydig细胞Atrn mRNA,Atrn蛋白表达量随培养天数的延长而减少,可能与睾酮的变化有关。
Objective: To investigate the expression of Attractin (Atrn) in cultured adult rat Leydig cells. Methods: The gene and protein expression of Atrn in Leydig cells was quantitated by Real - time polymerase chain reaction (PCR) and western blot methods, respectively, while testosterone in the medium was detected by Chemiluminescence Immunoassay. Results: Expression levels of gene and protein of Atrn and the level of testosterone was decreased during culturing process. Multiple correlation analysis showed a positive correlation between the gene and protein expression levels of Atrn(r =0. 953, P 〈0.05), and a positive correlation between the protein expression levels of Atrn and level of testosterone(r = 0. 976, P 〈 0.01 ). Conclusion : Expression level of Atrn is decreased in cultured Leydig cells, which may be related with testosterone production.
